摘要: Transcription factors mediate the formation of nucleoprotein complexes that are critical for efficient regulation epigenetic switches. In these complexes, DNA is frequently bent or looped by protein; other times, strong interactions lead to fully wrap regulatory protein(s). The equilibrium between bending, looping, full and partial wrapping governs level transcriptional tuned biophysical parameters. Characterization structure, kinetics, thermodynamics such fundamental understanding molecular mechanisms underlie operation genetic switches controlled them. Here, we describe in detail how perform tethered particle motion experiments aimed at protein-DNA influence breakdown complexes.
