Gemcitabine Functions Epigenetically by Inhibiting Repair Mediated DNA Demethylation

摘要: Gemcitabine is a cytotoxic cytidine analog, which widely used in anti-cancer therapy. One mechanism by gemcitabine acts inhibiting nucleotide excision repair (NER). Recently NER was implicated Gadd45 mediated DNA demethylation and epigenetic gene activation. Here we analyzed the effect of on demethylation. We find that inhibits specifically Gadd45a reporter activation demethylation, similar to topoisomerase I inhibitor camptothecin, also NER. In contrast, base inhibitors had no Xenopus oocytes, synthesis accompanying oct4. mammalian cells, induces hypermethylation silencing MLH1. The results indicate Thus, can function epigenetically provides tool manipulate methylation.
