Magnetic nanoparticle based purification and enzyme-linked immunosorbent assay using monoclonal antibody against enrofloxacin

摘要: Monoclonal anti-enrofloxacin antibody was prepared for a direct competitive enzyme-linked immunosorbent assay (ELISA) and purification system using monoclonal (mAb) coupled magnetic nanoparticles (MNPs). The IC50 values of the developed mAb enrofloxacin (ENR), ciprofloxacin, difloxacin, sarafloxacin, pefloxacin, norfloxacin were 5.0, 8.3, 9.7, 21.7, 36.0, 63.7 ng/mL, respectively. lowest detectable level ENR 0.7 ng/mL in ELISA system. To validate food matrix, known amounts spiked meat egg samples at 10, 20 30 ng/mL. Recoveries ranged from 72.9 to 113.16% with coefficient variation (CV) 2.42 10.11%. applicability mAb-MNP verified by testing recoveries residue three different matrices. 75.16 86.36%, while CV 5.08 11.53%. Overall, ENR-specific use ELISAs detection animal samples. Furthermore, we suggest that mAb-coupled MNPs could be useful determination food.
