摘要: ABSTRACT The acute effects of inorganic lead salts on mammalian cells in vitro were investigated using L-A cells, a subline L 929 mouse fibroblasts, HeLa-cells, human portio-carcinoma line, and diploid fibroblasts. Lead caused growth inhibition which was independant the anion employed could be prevented by addition Ca-EDTA. LC50 approximately 1 mM when exposing for 2 days × 10−4 M treating then 7 days. release lactic acid dehydrogenase not elevated indicating lack gross membrane damage lead. Concomitant with dose-dependent proliferation fraction synthesizing DNA mitotic rate lowered; cytophotometry block G1 phase cell cycle demonstrated. Energy metabolism appeared affected as evidenced rise lactate production. In HeLa fibroblasts aberrations observed. Under continuous exposure developed resistance, even to concentrations range LD50. kinetics acquirement resistance are resistant adjusted more quickly higher doses than controls. Low-resistance strains retained characteristics parent but increasing levels tolerance multiplication rates declined significantly. After ca. 60 generations heavy metal-free environment had major degree lost their that induced is adaptation rather mutation selection.
