摘要: Flavonoids may be a principal contributor to the cancer preventative activity of fruit- and vegetable-rich diets there is interest in their use as dietary supplements. However, potential conflict between cytoprotective cytotoxic activities flavonoids, efficacy anti-cancer agents unresolved. Here, integrity survival HL-60 promyelocytic leukaemia cells following short-term (90 min) exposure abundant flavonoid kaempferol (1-100 microM) reported. Supplementation initially decreased reactive oxygen levels but, paradoxically, dose-dependent increase single-strand DNA breakage occurred. was no oxidised purines or membrane damage. Following 24-h recovery period non-kaempferol supplemented media, had declined exposed control cultures possessed similar levels. A reduction (3)H-thymidine incorporation occurred with > =10 microM kaempferol. One hundred micromolar kaempefrol increased proportion G(2)-M phase, sub-G(1) content enhanced 'active' caspase-3 expression but only induced loss mitochondrial within minority cells. The relevance damage non-overtly oxidatively stressed environment disease therapeutic discussed.
