摘要: Top of pageAbstract Branched polyethylenimine 25,000 is an efficient cationic polymeric transfection agent that possesses both DNA condensation and endosome lytic functionalities. Its abundant primary, secondary tertiary amine groups contribute to activity as well significant toxicity cells. To better understand what extend each type group responsible for toxicity, selective chemical modification was performed on individual or types PEI by acylation (charge elimination) reductive amination (transformation amines). The resulted derivatives have different amounts varied degrees charge density. Here we show the synthesis these characterization in relationship binding, efficiency, cytotoxicity. Our initial results suggest primary are mainly high affinity binding elimination a portion polymers with enhanced gene efficiency significantly reduced toxicity. Secondary also contributed certain degree while contain entirety still transfectionally competent. These add existing knowledge about structural details PEI25,000 relevant its biological activities.
