摘要: Monocytes play a crucial role in controlling malaria infection. To facilitate our research into the development of antibody-mediated immunity against pregnancy-associated we have established several novel malaria-specific flow cytometric phagocytosis assays based on ethidium bromide staining DNA present blood stage trophozoites. The first assay quantifies ability sera to opsonise trophozoites and promotes by monocytic cell line THP1. This measures levels functional antibodies chosen strain parasite. second is whole which phagocytic patient monocytes ex vivo. third employs simultaneous labelling with erythrocytes fluorescein isothiocyanate compare both non-infected parasitised assess possible bystander effects uninfected erythrocytes. These advantage over other that they are rapid, simple specific malaria-infected cells avoid potential bias associated manual counting.
