G protein-coupled receptors (GPCRs) form large group of with vast array critical functions, and direct indirect interactions are thought to control various essential aspects receptor function. presence cholesterol, in particular, been focus number recent studies, varying receptor-dependent effects reported. However, possibility specific cholesterol binding sites on GPCRs remains debatable at present. A deeper structural mechanistic understanding complex delicately balanced nature GPCR–bilayer only revealed so far studies non-ligand binding, class GPCR, rhodopsin. Further investigations if we appreciate fully role composition GPCR activation signalling; indeed, improvements expression purification, along development novel reconstitution methods should make these types biophysical much more accessible. In this review highlight latest research GPCR–membrane some tools available for detailed studies.
