摘要: Due to increasing concerns about the use of antibiotic growth promoters (AGP) in livestock production and their complete ban European Union 2006, suitable alternatives are urgently needed. Among others, anti-inflammatory activities AGP discussed as putative mode action. As numerous phytochemicals known modulate cellular antioxidant capacity immune response, we studied antioxidative properties a phytogenic (plant-derived) feed additive (PFA) intestinal porcine epithelial cells (IPEC-J2). The effects PFA were compared with those selected ingredients (grape seed extract [GRS], licorice [LIC], menthol [MENT], methyl salicylate [MES], oak bark [OAK], oregano essential oil [ORE], plant powder mix [PLA]), tylosin (TYL). Oxidative or inflammatory stress was induced by stimulating IPEC-J2 hydrogen peroxide (H2O2; 0.5 mM) tumor necrosis factor alpha (TNF-α; 10 ng/mL), respectively. additives assessed reactive oxygen species (ROS)-sensitive probe measuring expression 6 target genes via quantitative real-time PCR (RT-qPCR). Anti-inflammatory potential analyzed using nuclear kappa-light-chain-enhancer activated B (NF-κB) reporter gene assay. Moreover, levels NF-κB measured RT-qPCR analysis, release IL-6 ELISA. Significant decreases ROS upon H2O2 treatment observed for (P < 0.001), LIC ORE 0.05), GRS 0.01). No significant changes found. activation TNF-α significantly inhibited 0.05) 0.001). reduced IL-8 C-C motif chemokine ligand 2 (CCL2; P well 0.05). other compounds TYL did not affect any parameters. In summary, revealed PFA, LIC, ORE, GRS, IPEC-J2, providing better understanding action this under our experimental conditions.
