摘要: Prenylation is a post-translational protein modification process that results in correct localization to membranes the cell. It mediated by at least three prenyltransferases (PTFs) eukaryotic cells. The RAM2 gene encodes common α-subunit of two PTFs, farnesyltransferase (FTase) and geranylgeranyltransferase I (GGTase I). In this study, prenylation pathway opportunistic pathogen Candida albicans was analysed. heterozygote trisomy test used demonstrate essential viability C. albicans. mRNA yeast hyphae growth states not detected Northern blot analysis, but RT-PCR. Drugs inhibit mammalian PTFs do alter cell growth, they FTase GGTase activities cell-free enzyme assays. data from genetic studies assays suggest drugs may have access cytoplasm. regulation PTF subunits also examined strain which under control regulable promoter. Overall, study demonstrated albicans, indicating an indispensable cellular yeast.
