摘要: The Mason-Pfizer monkey virus (MPMV) genome contains a cis-acting element that serves to facilitate nucleocytoplasmic export of intron-containing RNA. This element, known as the constitutive transport (CTE), has been mapped 154-nt region close 3' end MPMV genome. CTE degenerate direct repeat approximately 70 nt. We have probed secondary structure using double-strand- and single-strand-specific ribonucleases chemical modification agents. A mutational analysis was also performed confirm critical features structure, well identify regions contain sequence-specific information required for function. Our results indicate forms long stem 9-nt terminal hairpin loop two identical 16-nt inner loops. sequences are rotated 180 degrees relative each other within structure. shows primary in important function, suggesting they may binding sites cellular proteins involved RNA export. Interestingly, with significant homology found genomes spumaviruses mouse intracisternal particles.
