Role of Penaeus monodon hemocyte homeostasis associated protein (PmHHAP) in regulation of caspase-mediated apoptosis.

摘要: The viral responsive protein, PmHHAP, plays an important role in the control of hemocyte homeostasis shrimps during infection. In this study, we further investigate PmHHAP regulation apoptosis. RNA interference (RNAi) mediated gene silencing was used to suppress expression and change apoptosis determined knockdown shrimp. Within circulating hemocytes, increased number annexin V-positive apoptotic cells combined caspase-3/-7 activity induced characteristic DNA ladder. Furthermore, down-regulation accompanied by significantly altered apoptosis-related proteins including effector caspases, PmCaspase PmCasp. Yeast two-hybrid co-immunoprecipitation assays showed that binds p20 domain Moreover, recombinant protein able reduce caspase actinomycin D-treated rPmCasp-treated cells. Taken together, our data indicate regulates inhibits cell death through activation.
