Ambient oxygen tension modulates endothelial fibrinolysis

摘要: Abstract Purpose:  Vascular procedures reoxygenate ischemic endothelial cells (EC) and arterialize saphenous vein (HSV) EC. The balance between the EC-derived fibrinolytic components, plasmingogen activator (tPA), plasminogen inhibitor (PAI-I) contributes to maintaining thromboresistance. This also affects proteolysis through plasmin generation, mediating matrix metabolism migration, angiogenesis, theoretically affecting development of intimal hyperplasia. Methods:  To explore impact varying oxygen tensions on EC fibrinolysis, HSV human umbilical (HUV) were subjected PO 2 40 mm Hg for 24 hours with restoration 150 hours. tPA PAI-1 antigen tPA/PAI-1 ratio in conditioned media (CM), expressed as ↑ or ↓ % change, normalized cell count, versus controls, analyzed by enzyme-linked immunosorbent assay. Cellular mRNAs assessed Northern analysis. Results:  The but not was significantly decreased after first HSVEC 48 both HUVEC when compared controls. Messenger RNA unchanged mRNA increased Hg, returning baseline within restoration. Conclusions:  These data support hypothesis a shift altered ambient O exposure endothelium demonstrate that are more sensitive tension than HUVEC. Altered depress fibrinolysis this model.
