摘要: Abstract Mutations in the suppressor of Hairy-wing [su(Hw)] locus reverse phenotype a number tissue-specific mutations caused by insertion gypsy retrotransposon. The su(Hw) gene encodes zinc finger protein which binds to 430 bp region shown be both necessary and sufficient for its mutagenic effects. causes inactivation enhancer elements only when binding is located between these regulatory sequences promoter. To understand molecular basis inactivation, we tested effects on expression mini-white gene. We find that stabilizes from chromosomal position-effects euchromatic locations inactivating negative positive present flanking DNA. Furthermore, partially protects transposon insertions heterochromatin. explain our current results, propose alters organization chromatin creating new boundary pre-existing domain higher order structure. This separates enhancers silencers distal into an independent unit activity, thereby causing their inactivation.
