Basal Level of Autophagy Is Increased in Aging Human Skin Fibroblasts In Vitro, but Not in Old Skin

摘要: Intracellular autophagy (AP) is a stress response that enhanced under conditions of limitation amino acids, growth factors and other nutrients, also when macromolecules become damaged, aggregated fibrillated. Aging generally accompanied by an increase in intracellular due to all the above factors. Therefore, we have compared basal levels AP serially passaged human facial skin fibroblasts undergoing aging replicative senescence vitro, ex vivo biopsies from photo-protected photo-exposed area arms 20 healthy persons young old ages. Immunofluorescence microscopy, employing antibodies against specific microtubule-associated protein-1 light chain-3 (LC3) as well established marker AP, showed 5-fold level LC3 near senescent fibroblasts. However, no such age-related fluorescence could be detected full thickness sections obtained 10 (age 25 30 yr) 60 65 donors. Furthermore, there was difference areas arm.
