摘要: Abstract Quantification of the effect antiretroviral drugs on insulin aggregation process is an important area research due to serious metabolic diseases observed in AIDS patients after prolonged treatment with these drugs. In this work, multivariate curve resolution alternating least squares (MCR-ALS) was applied infrared monitoring presence three quantify their effect. To evidence concentration dependence process, mixtures at two different insulin:drug molar ratios were used. The interaction between and each drug analysed by 1H NMR spectroscopy. all cases, monitored during 45 min aggregates further characterised scanning electron microscopy (SEM). MCR-ALS provided spectral profiles insulin–drug conformations that are involved process. Their feasible band boundaries calculated using MCR-BANDS methodology. kinetic describe pathway characterise involved. retrieved results show modifies conformation a way, promoting formation aggregates. Ritonavir shows strongest promotion aggregation, followed efavirenz zidovudine. studied range, only for zidovudine, shorter time obtained as amount zidovudine increased. This factor also affected pathway.
