摘要: Publisher Summary This chapter discusses photoaffinity substrate analogs for eukaryotic prenyltransferase. The technology has been exploited identifying the catalytic sites of several prenyltransferases (dimethylallyltransferases). Two independent and different approaches have taken. In Allen's laboratory an ingenious procedure is utilized to prepare analogs. A photoreactive moiety attached end isoprenoid chain. general can be applied almost any polyisoprenoid molecule that a terminal isopropylidene group. One had found inhibitory phenylethyl pyrophosphate. suggested aryl azide bearing ethyl pyrophosphate provides useful photolabel this enzyme, it was following are synthesized. Concentrations esters determined by analysis total phosphate as described.
