Insulin-like effects of trypsin on fat cells. Localization of the metabolic steps and the cellular site affected by the enzyme.

摘要: Abstract The mechanism of the insulin-like action trypsin was studied. When fat cells were exposed to (1 mg per ml) for 15 sec, glucose metabolism in stimulated. This stimulation (like that by insulin) inhibited phloretin or 3-O-methyl-d-glucose. By above treatment with trypsin, cellular lipolysis induced epinephrine blocked a large extent, but caffeine only partially inhibited. These effects on similar those insulin at low concentration (20 30 microunits ml). Trypsin did not inhibit N6, O2'-dibutyryl adenosine 3',5'-monophosphate. treated incubated buffer few hours after inactivation enzyme, had recovered from enzyme. recovery significantly cycloheximide. "recovered" less sensitive both and as compared untreated cells. decrease sensitivity prevented when dose added incubation medium prior treatment. data consistent an assumption (a) interacts initially receptor site cells, (b) activates produce metabolic responses characteristic insulin, (c) eventually modifies render than normal trypsin.
