The utilization of RAPD-PCR for identifying thermophilic and mesophilic Bacillus species.

摘要: A random amplified polymorphic DNA fingerprinting assay has been optimized that is able to discriminate between numerous thermophilic and mesophilic bacillus species strains. Included in the analyses are (able grow at 55°C) strains of Bacillus stearothermophilus, B. kaustophilus, coagulans, sphaericus, thermodenitrificans, thermocatenulatus, thermoleovorans, licheniformis, brevis, thermoglucosidasius, caldolyticus, caldotenax, caldovelox, thermocloacae smithii. Mesophilic pumilus, subtilis, megaterium, circulans, cereus mycoides can also be used for with assay. Increasing concentration primer from 0.2 2.0 μM shown have a significant effect on increasing number amplification products discrimination or identification individual species. It suggested this may general way improving resolution RAPD protocol. The conditions successfully trace licheniformis other an industrial setting.
