Variability of 6-mercaptopurine pharmacokinetics during oral maintenance therapy of children with acute leukemia

摘要: The effects of some environmental and genetic factors on the inter- intraindividual variations 6-mercaptopurine (6-MP) pharmacokinetics were studied in children oral remission maintenance therapy for acute lymphoblastic leukemia or non-Hodgkin’s lymphoma. Blood samples obtained 0–4 h after drug intake. 6-MP concentrations determined plasma erythrocyte concentrates. influence food was examined a prospective study 15 children. Each child four times, twice fasted state intake standardized, milky, breakfast. There pronounced variations. Food seemed to reduce these but there no significant changes peak area under concentration vs time curves (AUC) between fed states. reduced both plasma, from 1.8 1.1 (P 70 mg m−2 had higher AUC fasting state, while five out eight patients receiving <70 state. cytochrome P-450-dependent hydroxylation capacity evaluated with debrisoquine correlation found 6-MP.
