摘要: pet18 mutations in Saccharomyces cerevisiae confer on the cell inability to maintain either L-A or M double-stranded RNAs (dsRNAs) at nonpermissive temperature. In vitro experiments, we examined effects of RNA-dependent RNA polymerase activity associated with virus-like particles (VLPs). caused thermolabile VLPs, and this thermolability was found be due instability VLP structure. The did not affect VLPs. Furthermore, temperature sensitivity wild-type differed substantially from that polymerase. From these results, other genetic biochemical lines evidence which suggest replication dsRNA requires presence dsRNA, propose primary effect mutation is structure mutants comes loss dsRNA.
