Half‐Life of Histone H4 mRNA in Xenopus laevis Embryonic Cells at Different Stages: (histone H4 mRNA/Northern blotting/Xenopus embryonic cells/ half‐life/actinomycin D)

摘要: By Northern blotting methods we have recently found that the relative amount of histone H4 mRNA per Xenopus embryo is constant during cleavage, doubles gastrula stage, then decreases at neurula stage to level slightly lower than cleavage-embryo. At each developmental will depend on three parameters: maternal remaining, rate synthesis and decay mRNA. In present experiment, compared rates in embryonic cells different stages by pulse-chase experiments with actinomycin D as an inhibitor transcription. Under conditions used, half-life was estimated be 90, 80, 100 min, for late blastula, stages, respectively. The a 90 min blastula predicts exhaustion most early gastruia stage. longer suggests more stable cells, therefore, large decrease observed does not increase turnover-rate Probably, synthesize less but utilize it time.
