Response of the vacuolar ATPase subunit E to RNA interference and four chemical pesticides in Leptinotarsa decemlineata (Say).

摘要: Vacuolar-type H(+)-ATPases (vATPases) are localized in the apical membranes of nearly all epithelial tissues insects, energize to absorb and/or secrete ions and fluids, play essential roles many physiological functions. Here we cloned characterized a 1041-bp full-length vATPase subunit E cDNA (named as LdATPaseE) that encoded 226-amino acid protein Leptinotarsa decemlineata. LdATPaseE mRNA levels were constantly increased from egg third- fourth-instar stages, dropped wandering pupal stages elevated again adult stage. It was highly expressed ileum rectum, moderately Malpighian tubules, midgut foregut, lowly fat body, ventral ganglion, epidermis haemocytes fourth instars. After continuously ingested double-stranded RNAs originated two fragments LdATPaseE1 LdATPaseE2, target larvae reduced by 85% 55%, larval growth survival significantly affected. Furthermore, topical application fipronil, butane-fipronil, endosulfan cypermethrin upregulated expression up 8.3, 4.2, 2.8 6.2-fold 1 day after experiment, 15.8, 3.4, 3.6 4.5-fold 2 days treatment. seems depletion is lethal, indicating targeting vATPases dsRNA appears promising means combating L. Moreover, pesticide inducible gene may role toxicity.
