Evaluation of bronchodialatory and antimicrobial activities of Otostegia fruticosa: A multi-mechanistic approach.

摘要: Abstract Otostegia fruticosa, a plant belonging to the family Lamiaceae, is endemic Ethiopia. In Ethiopian traditional medicine, O. fruticosa has been used for treatment of several respiratory-related disorders. The present study was designed evaluate bronchodilatory and antimicrobial activities leaves crude extract (Of.Cr). Ex-vivo experiments were conducted on guinea-pig trachea provided with physiological oxygenated buffer solution using emkaBath setup. analyzed by gas chromatography-mass spectrometry. Of.Cr, showed presence terpenes, fragrance components, saponins, higher fatty acids. Of.Cr when tested contracted tracheal chains carbamylcholine (CCh, 1 µM) high K+ (80 mM) produced relaxation showing potency against CCh incomplete inhibition K+. Dicyclomine, as positive control, also selectively inhibit compared its effect anticholinergic curves, at 1 mg/mL deflected CCh-induced concentration–response curves (CRCs) competitively right like dicyclomine (0.03 µM) atropine whereas dose (3 mg/mL) non-parallel shift in (0.1 µM). calcium channel inhibitory assay, 3 & 5 mg/mL, CRCs Ca++ verapamil, control. concentrations (1–3 mg/mL) increases cAMP levels isolated homogenates, similar control phosphodiesterase inhibitor (papaverine). When antibacterial activity standard clinical strains, found more active (MIC 475 µg/ml) S. aureus (NCTC 6571), while maximum 625 µg/ml) observed MRSA. These results determine mechanistic pathways combination dual voltage-gated channels.
