Use of tRNA Gene Probes to Identify Polymorphic Loci in the Bovine Genome

摘要: A 30-mer oligonucleotide probe encoding the "A box" and anticodon loop regions of a human glycine tRNA gene was used to isolate 581bp DNA fragment from bovine genomic library. Although cross-hybridizing segment found not encode any or pseudogene, region with homology "C-element" "BOV-tA" type Alulike artiodactyl retroposons identified. This cross-hybridization determined be result conserved RNA polymerase III promoter elements in portion these repetitive elements. microsatellite repeat (TC) also associated this element. Future screening for genes will require use a) longer probes higher stringency hybridization conditions b) simultaneous 5' 3' ends which avoid Pol boxes.
