摘要: Membrane-bound proteins in plasma membrane enriched fractions from cultured rat RPE were analyzed by two-dimensional gel electrophoresis. Membrane characterized on three increasingly specific levels. Total protein was visualized silver staining. A maximum of 102 separate counted silver-stained gels. Glycoproteins labeled with 3H-glucosamine or 3H-fucose and detected autoradiography. Thirty-eight fucose-labeled 61-71 glucosamine-labeled identified. All the glucosamine-derived radioactivity. Proteins exposed at cell surface lactoperoxidase-catalyzed radioiodination prior to preparation membranes for analysis. Forty 125I-labeled resolved electrophoresis/autoradiography. Comparison glycoprotein map showed that a number these glycoproteins. Two-dimensional maps total protein, fucose-labeled, glycoproteins, dystrophic (RCS rdy-p+) normal (Long Evans RCS rdy+p+) compared. No differences surface-labeled observed. However, results suggest 183K is more heavily glycosylated glucosamine fucose as compared RPE.
