摘要: Amelotin (AMTN) is induced upon initiation of apoptosis by transforming growth factor beta1 (TGFβ1) and mediated Smad3 in gingival epithelial cells (GE1 cells). This upregulation AMTN gene expression temporary, the mechanism responsible still unclear. The present study investigated transcriptional downregulation TGFβ1-induced GE1 during progression apoptosis. To examine time-dependent changes levels AMTN, Bax mRNA TGFβ1, real-time PCR analyses were performed. Immunocytochemistry was carried out to detect Bax. Transient transfection performed using mouse promoter constructs various lengths including Smad response elements (SBEs), presence or absence TGFβ1. Changes binding SBEs resulting from overexpression examined ChIP assays. Overexpression dramatically downregulated transcription gene. this inhibited overexpression. These results show that TGFβ1 are decreased robust cells.
