摘要: The effects of short-term oral administration red wine polyphenolic compounds (RWPCs) on blood pressure and vascular reactivity were investigated in rats. consequence RWPCs treatment agonist-induced contractility rat aorta with respect to Ca2+ handling was assessed, by examining both intracellular store extracellular influx components the response. Rats treated daily for 7 days intragastric either 5% glucose, or (20 mg/kg) [from two different sources, i.e. Provinols (SFD, Vallont Pont d'Arc, France) RWPC1 (INRA, Montpellier, France)]. Administration these produced a decrease systolic pressure. smooth muscle without endothelium exposed noradrenaline. In Ca2+-free medium, but not induced an increase noradrenaline-induced contraction. After depletion stores noradrenaline addition CaCl2 continuous presence agonist contraction, which significantly between control, Provinols- RWPC-treated inhibitor sarcoplasmic reticulum Ca2+-ATPase, thapsigargin, reduced contraction medium RWPCs-treated aorta, as compared that control. Interestingly, inhibitory effect thapsigargin response linked release from internal vessels completely prevented NO-synthase inhibitor, L-nitro arginine methyl ester, guanylyl cyclase, oxadiazoloquinoxaline protein kinase G 8-Bromoguanosine-3′-5-cyclic monophosphorothioate, Rp isomer. These results suggest rats subtle alteration thapsigargin-sensitive component store. Calcium sensitive implicated this mechanism. prevention inhibitors NO/cyclic guanosine monophosphate pathway after highlights role NO phenomenon.
