摘要: We report here on the use of RNA interference (RNAi) to create pupal and adult loss-of-function phenotypes in red flour beetle, Tribolium castaneum, by injection double-stranded (dsRNA) into late instar larvae (we refer this method as larval RNAi). RNAi is well-established a useful mimic many organisms including insects. However, with few exceptions (such fruit fly Drosophila melanogaster), analysis has usually been limited studies embryogenesis. Here we demonstrate that green fluorescent protein (GFP) dsRNA body cavity can inhibit GFP expression beginning shortly after continuing through stages. Tc-achaete-scute-homolog (Tc-ASH) revealed induce morphological defects beetles, also affects entire rather than being localized near site injection. The technique will be analyze gene functions post-embryonic development, giving us opportunity study molecular basis diversity various organisms.
