摘要: Objectives The main aim of this work is to study how E5, E6 and E7 oncogenes human papillomaviruses type 18 could cooperate among each other boost key cancer cell features such as uncontrolled proliferation, enhanced migration capacity, invasion, relates with oxidative stress. Methods We generated three HaCaT lines, that are spontaneously immortalized, expressing the following combination oncogenes: E5–18, E6/E7–18 E5/E6/E7–18 non-transduced cells a control. Cell proliferation was assessed using MTT assay. Cellular studied wound healing assay by scratching monolayer in order generate “wound”. Invasiveness potential transwell collagen invasion Intracellular oxidants production four lines measured fluorescent probe CM-H2DCFDA. Catalase activity assayed spectrophotometrically, decomposition 10 mM H2O2 catalase contained samples at 240 nm. Total glutathione lysates quantified HPLC, PRX1 expression levels Western Blot. Results showed statistically significant increment E5/E6/E7 respect lines. Similar results were obtained assay, assays. oncoproteins involved ROS metabolism observed diminished but augmented significantly GSH PRX1. Conclusions This demonstrates together augment malignant transformation cells.
