摘要: Immunoscreening of a Candida albicans expression library resulted in the isolation novel gene encoding 32.9-kDa polypeptide (288 amino acids), with 27.7% homology to product Saccharomyces cerevisiae YGR106c, putative vacuolar protein. Heterozygous mutants this displayed an altered budding growth pattern, characterized by formation chains buds, decreasingly size towards apex, without separation daughter buds. Consequently, was designated ABG1. A conditional mutant for ABG1 remaining allele under control MET3 promoter did not grow presence methionine and cysteine, demonstrating that essential viability. Western analysis revealed major band, mainly particulate fraction (P40) enriched vacuoles, tagging green fluorescent protein confirmed Abg1p localized vacuole. Vacuole inheritance has been linked regulation branching frequency C. albicans. Under repressing conditions, had increased hyphal inducing conditions sensitivity substances interfered cell wall assembly. Repression strain caused disturbance normal number vacuoles both yeast mycelial cells also asymmetric vacuole associated characteristic pattern germ tubes These observations indicate plays key role biogenesis, cytokinesis, branching.
