摘要: We have investigated in detail the cleavage of human high molecular weight (HMW) kininogen by plasma kallikrein and revealed formation a nicked novel kinin-free protein (KFP) as intermediate products. The single chain HMW (Mr=120,000) was three-step reaction. first yielded composed two disulfide-linked 62,000 56,000 daltons chains. second kinin an protein, KFP-I, which apparently equal size to kininogen. third stable KFP-II, 45,000 liberation 8,000 fragment identified when isolated SP-Sephadex C-50 chromatography reduced alkylated KFP-I cleaved into chain. Although antiserum against cross-reacted with low (LMW) kininogen, specific for These results suggest that antigenic determinant groups common LMW kininogens are located heavy chain, while those light is known retain blood coagulation activity.
