摘要: The present study is aimed to examine the shape effect on lineage commitment of stem cells in growth medium free external chemical induction factors. Aspect ratios (ARs) were controlled by micropatterns with cell-adhesive microislands AR 1, 2 and 8 potent nonfouling background poly(ethylene glycol) hydrogels, single well shaped for 19 days. Mesenchymal (MSCs) derived from rat bone marrow cultured osteogenic medium, adipogenic mixed coinduction also medium; alkaline phosphatase (ALP) oil droplets employed as indicators osteoblasts adipocytes, respectively. Those observed all three media early day 7, at a longer culture time till 13. While significant monotonic decrease adipogenesis was increase AR, non-monotonic change osteogenesis found optimal about 2. relative gene expressions further verified above findings. As result, cell itself an inherent cue regulate differentiation, let alone or without Such disappeared upon addition microfilament inhibitor cytochalasin D Rho-associated protein kinase (ROCK) Y-27632. So, formation cytoskeleton necessary effect, ROCK-pathway-related tension responsible even medium.
