Alteration of intracellular metabolome in osteosarcoma stem cells revealed by liquid chromatography-tandem mass spectrometry

摘要: Cancer stem cells (CSCs) are the origin of many malignant tumours, including osteosarcoma that mainly affects adolescents and is accompanied by a poor prognosis. However, little known about intrinsic biological information cells, particularly for metabolomics features. Hence, an ultra-high performance liquid chromatography coupled with tandem Q-Exactive Orbitrap mass spectrometer (UHPLC-QE-MS)-based approach was developed to investigate metabolism changes in human (HOS) cell line order understand its possible mechanism. HMDB, METLIN m/z Cloud databases were used identify metabolic markers. Additionally, compounds further identified using standards metabolites. Comparing HOS-CSCs non-CSCs, 154 different metabolites both positive negative modes. Based on clearly distinct metabolites, changed pathways analysed MetaboAnalyst. The top five altered included alanine, aspartate glutamate metabolism; arginine proline glutathione cysteine methionine citrate cycle (TCA cycle). downregulation TCA elevation oxidized levels suggested decline mitochondrial metabolism, while most amino acid metabolisms upregulated. Further experiments analysis activity confirmed above hypotheses deduced from results. These findings not only enhance our understanding metabolome but also demonstrate great potential such method based UHPLC-QE-MS large-scale studies.
