摘要: A new human thyroid carcinoma cell line, KTC-1, was established from the malignant pleural effusion of a recurrent patient. Cytogenetic analysis revealed normal karyotype, and no p53 mutation in exons 5‐9 detected. This line is tumorigenic athymic nude mice. Histological findings by light electron microscopy, such as absence follicular structures existence intranuclear cytoplasmic inclusions psammoma bodies, indicated transplanted tumors to be poorly differentiated papillary carcinoma. low expression level thyroglobulin detected immunocytochemistry RT-PCR. Messenger ribonucleic acid (mRNA) transcription factor-1 PAX-8 also No mRNA TSH receptors, peroxidase, or Na 1 /I 2 symporter Interleukin-6 leukemia inhibitory factor were secreted into medium. These suggest this functionally differentiated. Moreover, all-trans-retinoic increased decreased both secretion interleukin-6 while significantly stimulating growth. RT-PCR retinoic receptors (RARs) that KTC-1 cells express moderate RARa -g, but RARb. may useful for studying redifferentiation therapy (J Clin Endocrinol Metab 85: 2889 ‐2896, 2000)
