摘要: Purpose: To investigate microRNA-181 (miR-181) as a potential therapeutic target in osteoarthritis (OA). Methods: MiR-181 expression was evaluated articular cartilage samples obtained from OA patients undergoing knee arthroplasty and non-OA (control) other orthopedic procedures. Following the isolation of total RNA, miRNA mRNA determined by real timepolymerase chain reaction (RT-PCR). Luciferase reporter assay mimic or inhibitor were then used to establish molecular miR-181 chondrocytes. Results: family members, namely, miR-181a, miR-181c miR-181d significantly upregulated compared control subjects. However, no significant difference up-regulation miR-181b expression. B-cell lymphoma 2 (BCL2), putative family, down-regulated Furthermore, luciferase confirmed direct interaction between miR-181a three prime untranslated region The 3’UTR BCL2 Transfection resulted suppression On hand, transfection led increased decreased interleukin 1-beta (IL1-β) induced apoptosis. Conclusion: is differentially expressed leads downregulation BCL2, regulator Therefore, may be treatment OA. Keywords: MicroRNA, Osteoarthritis, Apoptosis, 2, Transfection, Chondrocytes
