摘要: Purpose. To develop and validate an HPLC method for the quantitation of clozapine its metabolite, norclozapine in human plasma, rat various plasma lipoprotein fractions. Methods. Using liquid-liquid extraction, clozapine, are extracted from biological matrix with MTBE. After concentration, residue was reconstituted 1mM HCl injected on to a C6 Phenyl column (3μm, 2.0×150 mm). Mobile phase 10mM Ammonium Acetate, pH 5—Acetonitrile—Methanol (5:3:2, v/v/v). Loxapine served as internal standard. Absorbance measured at 254 nm. Results. Quantitation limits 100 ng/mL 50 ng/mL, respectively. Recovery both near 100%. Percent accuracy intraday variability TRL, HDL, LDL, LPDP fraction between 92 113% analytes.
