摘要: RNA interference (RNAi) is an evolutionarily ancient method of genome defense in many organisms. RNAi triggered when a cell encounters long double-stranded (dsRNA) and results the silencing gene expression (Fire et al., 1998). The dsRNA processed by RNAse III-like nuclease, Dicer, into 20–25nt duplex RNA, termed short interfering (siRNA, Zamore 2000; Hammond 2000). siRNAs assemble RNA-induced complexes (RISCs) containing endoribonucleases are unwound. It believed that sense strands degraded, while anti-sense activate RISC to participate repeated cycles cleavage degradation complementary cognate mRNA, effectively such no protein made. For this reason, seen as useful tool for analyzing function. Given introducing mammalian cells initiates potent antiviral response (Stark 1998), most investigators bypass or expressing siRNAs, which key mediators (Elbashir 2001a b).
