摘要: RNA interference (RNAi) is an evolutionarily ancient method of genome defense in many organisms. RNAi triggered when a cell encounters long double-stranded (dsRNA) and results the silencing gene expression (Fire et al., 1998). The dsRNA processed by RNAse III-like nuclease, Dicer, into 20–25nt duplex RNA, termed short interfering (siRNA, Zamore 2000; Hammond 2000). siRNAs assemble RNA-induced complexes (RISCs) containing endoribonucleases are unwound. It believed that sense strands degraded, while anti-sense activate RISC to participate repeated cycles cleavage degradation complementary cognate mRNA, effectively such no protein made.
