Mutational analysis of the PTEN gene localized at chromosome 10q23 in Thai patients with gliomas

摘要: Phosphatase and tensin homologue deleted on chromosome 10 (PTEN), a tumor suppressor gene located 10q23, has recently been shown to act as phosphatidylinositol 3,4,5- triphosphate phosphatase modulate cell growth apoptosis. Genetic alterations in the PTEN occur several types of human cancers including glioblastoma, prostate, breast. We examined genetic gliomas. screened 28 astrocytic tumors for mutations throughout coding regions using polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP), Klenow treatment direct DNA sequencing aberrantly migrating PCR products. found nucleotide changes introns but no that caused amino acid alterations. Interestingly, intronic accompanying silent were only patients with glioblastomas. In conclusion, majority do not carry gene, which suggests there may be other genes are important tumorigenesis
