Cytotoxicity of apigenin on leukemia cell lines: implications for prevention and therapy

摘要: Natural-food-based compounds show substantial promise for prevention and biotherapy of cancers including leukemia. In general, their mechanism action remains unclear, hampering rational use these compounds. Herein we that the common dietary flavonoid apigenin has anticancer activity, but also may decrease chemotherapy sensitivity, depending on cell type. We analyzed molecular consequences treatment in two types leukemia, myeloid erythroid subtypes. Apigenin blocked proliferation both lineages through cell-cycle arrest G2/M phase HL60 G0/G1 TF1 cells. lines JAK/STAT pathway was one major targets apigenin. inhibited PI3K/PKB induced caspase-dependent apoptosis. contrast, no apoptosis detected cells, initiation autophagy observed. The block cycle induction observed this erythroleukemia line resulted a reduced susceptibility toward commonly used therapeutic agent vincristine. Thus, study shows although is potential chemopreventive due to leukemia arrest, caution intake should be taken during disease as it potentially interferes with cancer treatment.
