摘要: Bleomycin, an antitumor antibiotic with specific activity against squamous cell tumors of the skin, was administered iv every 4 days for 11 or 31 treatments to 14 dogs (0.156–5 mg/kg/day) and monkeys (0.5–8 mg/kg/day). Severe doserelated cutaneous toxicity included dermatitis, ulceration at pressure friction sites, onychoptosis focal alopecia. Foot pads housed in wire floor cages were more severely affected than those on solid floors. In dogs, lesions occurred only footpads tail tip, while neck, elbows, cubital fossa, ischial callosities palmar plantar surfaces hands feet affected. Microscopic changes sites ulceration, necrosis, subacute inflammation, hemorrhage, edema occasional foreign body granulomas. Collagen bundles both non-pressure showed increased homogeneity, but elastic reticular fibers no evident abnormalities. Atrophy sweat glands pilosebaceous apparatus also observed. Previously reported depression serum zinc concentrations correlated onset skin lesions, suggested that deficiency played a role pathogenesis lesions. Integumentary good visual index bleomycin toxicity.
