摘要: A confocal microscope can be considered as a 3D sampling instrument for collecting data from spatial structures, especially biological ones. Optimal collection in microscopes requires the adaptation of dimensions volume to lateral and axial raster parameters employed during collection. It is shown how, principle, partly manipulated by use variable pinholes both illumination detection paths. The effective spot will depend on optics used, degree aberration, alignment instrument. Measurements response fluorescence reflection some high N.A. lens systems function above factors are presented. computer-controlled instruments discussed, relation operation fluorescence. indicated that proper interpretation processing at least approximate knowledge applicable function.
