摘要: The application of a rapid and direct proteotyping approach with which to identify the gene origin viral antigens in reassortant influenza strain is demonstrated. strain, constructed for vaccine against type A 2009 H1N1 pandemic influenza, contains genes derived from wild-type (A/California/7/2009) an egg adapted high-growth (denoted NYMC X-157) earlier A/Puerto Rico/8/34 strain. employs modern proteomics methods high resolution mass spectrometry correctly establish that surface antigens, hemagglutinin neuraminidase, are A/California/7/2009 while those nucleoprotein matrix protein M1 X-157 This achieved both gel-separated whole digest. Furthermore, signature peptides detected spectra digested enable engineered be identified as virus subtype accord studies. results demonstrate provides more over RT-PCR characterize strains at molecular level. Given these pose greatest risk human animal health have been responsible all pandemics 20th 21st centuries, there vital need origins evolutionary history rapidly established.
