摘要: We have studied factor VII activation by measuring the ratio of clotting to coupled amidolytic activity (VIIc/VIIam) and cleavage 125I-factor VII. In purified systems, a low concentration Xa or higher IXa rapidly activated VII, yielding VIIc/VIIam 25 similar gel profiles heavy light chain peaks VIIa. On further incubation, VIIa diminished third 125I-peak appeared. When normal blood containing added 125I- was clotted in glass tube, rose fivefold, 20% cleaved. Clotting plasma an partial thromboplastin time (APTT) system yielded over 90% XII-deficient preincubated with antibodies X APTT XIa 19 about 60% cleavage, which indicates that IXa, at achievable plasma, can effectively activate undiluted tissue 15 20 whereas diluted only minimally. conclude both function as significant activators vitro mixtures but believe small amounts may be vivo during hemostasis.
