摘要: Eryptosis, the suicidal death of erythrocytes, is characterized by cell shrinkage and membrane scrambling with phosphatidylserine exposure at erythrocyte surface. Eryptosis triggered several stress conditions including isotonic (Cl(-) removal) energy depletion (glucose removal). Both are effective through an increase in cytosolic Ca(2+) concentration. Phosphatidylserine-exposing erythrocytes cleared from circulating blood. Enhanced eryptosis thus leads to anemia. Accordingly, drugs interfering may prove useful treatment The present study explored, whether caffeine interferes eryptosis. Erythrocyte was estimated annexin V-binding, volume forward scatter activity Fluo3 fluorescence. Under control conditions, affected less than 5% not significantly modified presence (50-500 microM). Glucose (for 48 hours) increased fluorescence V-binding decreased scatter, effects partially reversed (500 Low Cl(-) solution exchanged gluconate for similarly again In conclusion, inhibits entry following glucose counteracts during depletion.
