摘要: Acrylamide (ACR) is a vinyl monomer with established human neurotoxic effects, which characterized by the accumulation of neurofilaments (NFs) in distal swellings large axons peripheral and central nervous systems. However, mechanisms neurotoxicity remain unclear. The objective to investigate neuroprotective effect calpeptin (CP) on ACR-induced neuropathy its mechanism. Female adult Wistar rats were randomly divided into four groups (control, CP, ACR, ACR + CP group). Control group received 0.9 % saline, 30 mg/kg intraperitoneal injection. In addition, also 200 µg/kg CP. Gait analysis hind limb splay measured weekly analyze neurobehavioral changes. calpain activity changes NFs protein levels spinal cord are determined. Compared control group, body weight decreased 11.3 (P < 0.01), while increased significantly 8.3 0.01) compared end 4th week; gait score both 167 100 it 25.1 group; distance 76.7 49.5 15.4 at 14.9 10.0 4.2 light NF (NF-L), medium (NF-M) heavy (NF-H) subunits 81.2 %, 263.6 22.6 supernatant tissue 28.4 96.6 10.6 NF-L, NF-M NF-H 29.1 45.9 9.8 group. present results suggested that can relieve decrease degradation. may be one neuropathy.
