NuSAP is degraded by APC/C–Cdh1 and its overexpression results in mitotic arrest dependent of its microtubules' affinity

摘要: Abstract Microtubule associated proteins are involved in regulation of microtubule dynamics. Its mutation and dysregulation result severe consequences such as mitotic block apoptosis. NuSAP has been reported a protein, depletion which by RNAi results spindle deficiency cytokinesis failure. However, its role cell cycle how protein is controlled during progression still remains unclear. Here we show that can be ubiquitinated degraded APC/C–hCdh1 E3 ligase. Evolutionally conserved KEN box functions the degron NuSAP. Overexpression induces arrest domain nuclear localization both required for to induce arrest. Furthermore, overexpression cells accumulation with bundling deficiency. Thus, our give evidence first time level tightly regulated APC/C ubiquitin ligase complex dependent affinity.
