Inhibition of polyoma virus middle T antigen-associated tyrosyl kinase activity by N-ethylmaleimide.

摘要: The middle T antigen (MT Ag) encoded by polyoma virus has an associated protein kinase activity which transfers a phosphoryl group from ATP or GTP to tyrosine residue on MT Ag in immunoprecipitates formed between virus-infected transformed cell extracts and serum animals bearing polyoma-induced tumors. Incubation of such polyoma-transformed prior immunoprecipitation with the sulfhydryl reagent, N-ethylmaleimide (NEM), resulted significant inhibition Ag-associated activity. Inactivation this enzyme NEM was found be dependent upon incubation pH, time incubation, concentration. ATP, GTP, ADP presence Mg2+ were decrease rate NEM-mediated inactivation activity, while CTP UTP did not detectably alter NEM. These results suggest that possesses at least one NEM-sensitive essential for phosphotransferase may present near catalytic site.
