CHEMICAL, BIOLOGICAL AND IMMUNOLOGICAL CHARACTERIZATION OF SEPARATED ELECTROPHORETIC COMPONENTS OF HUMAN CHORIONIC SOMATOMAMMOTROPIN (HCS)

摘要: Purified HCS preparations often show heterogeneity when assayed by electrophoresis, although they may be considered homogeneous according to other criteria (N-terminal amino acid analysis, gel filtration and ultracentrifugation patterns, etc.). Using DEAE-cellulose chromatography, the two main components corresponding various degrees of hormone amidation were separated. The deamidation kinetic electrophoretically slower migrating component was studied found sensitive temperature and, a lesser extent, pH. Denaturing agents strongly increased rate, while no effect raising ionic strength. No difference evident between parent compared (on weight basis) in specific radioimmunoassay system. Even precipitin reaction agar with an anti-human growth (HGH) serum did not variations cross-reactivity versus HGH. Moreover, groups guinea pigs immunized single components, gave antibodies reacting identically HCS. prolactin-like activity differed determined pigeon crop-sac assay: proved less active than more deamidated one. On hand, different radioreceptor assays (employing membranes from rabbit mammary gland rat ventral prostate) greater for faster component.
