Improved quantitation of plasma lipids by direct gas-liquid chromatography

摘要: A preliminary digestion of total plasma lipid extracts with phospholipase C, which converts the lysolecithins, lecithins, and sphingomyelins into monoglycerides, diglycerides, ceramides, respectively, has been shown to facilitate subsequent determination lipids by gas-liquid chromatography. further improvement in chromatographic elution pattern results from acetylation or trimethylsilylation liberated alcohol moieties prior injection chromatograph. If tridecanoin is used as internal standard, quantitative estimates can be rapidly obtained for free cholesterol, sphingomyelins, cholesteryl esters, triglycerides, well fatty acids. Other do not occur sufficiently high concentrations interfere analysis. The requires 0.1-0.5 ml about 6 hr processing, but many samples processed at a time.
